CONSERVATION LABORATORY TECHNICIAN

This is a part-time position funded for 1 year. Expected hours: 20-29 per week

JOB FUNCTION

The Atlanta Botanical Garden invites applications for the position of Conservation Laboratory Technician, for the Southeastern Center of Conservation within the Department of Conservation & Research at the Atlanta Botanical Garden. The successful candidate will work under the direction of the Conservation Laboratories Supervisor. The primary duties of this position are to:
Perform seed and pollen banking, leaf tissue collection, and herbarium voucher preparation necessary to complete the objectives of the Institute of Museum and Library Services funded project, “Safeguarding the Atlanta Botanical Garden’s Conservation Collections”
Assist with DNA isolation, PCR, sample preparation, and general organization in the Conservation Genetics Laboratory.

ESSENTIAL DUTIES AND RESPONSIBILITIES:
Greenhouse and nursery collection of seed, pollen, leaf tissue, and herbarium vouchers. Seasonal travel to Gainesville, GA will be required for collections.
Perform laboratory functions such as DNA isolation using the CTAB method, PCR, leaf tissue preservation, seed counting, seed viability testing (tetrazolium assays, microCT scans, and X ray scans), labeling, and general organization.
Prepare samples for submission to external laboratories.
Maintain detailed records of all collections and laboratory procedures according to standard operating procedures.

QUALIFICATIONS:
Must be organized, detail-oriented, and adaptable.
Associate or Bachelor of Science degree in Biology, Botany, Biotechnology, Horticulture, or related field preferred.
Minimum of two years of technical laboratory experience.
Prior experience with seed and pollen collection, tetrazolium testing, DNA isolation, and herbarium voucher preparation is highly desired.
Must have a valid Driver’s License with good driving record
Must be able to lift and carry up to 50 lb.

PHYSICAL REQUIREMENTS: Ability to spend long periods of time in the laboratory performing complex protocols. Ability to take careful notes and improve laboratory organization. Ability to lift 50 lb. is required.

MENTAL REQUIREMENTS: Excellent organizational and time management skills. Ability to effectively respond to multiple, changing and often conflicting priorities is essential. Must be able to effectively interact with the general public and co-workers and peer professionals.

Katharine H. Putnam Fellowships in Plant Science

The Arnold Arboretum of Harvard University invites applicants for research fellowships in plant science. Putnam Fellowships offer excellent opportunities for advanced research and study using the Arboretum's living collections of woody plants. Scientists with a PhD and who have identified an independent research project that would utilize the Arboretum's living collections are encouraged to apply. The living collection, numbering some 15,000 plants, in over 2,200 species, is distinguished as one of the most thoroughly documented collections of temperate woody plants in the world. Taxonomic diversity and breadth within the collection are noteworthy, and the floras of China, Japan, and Korea are particularly well represented.

Proposals are sought from early-career individuals with a PhD in plant biology, evolution, plant genetics, plant ecology, horticulture, or related discipline. Applicants should be well positioned to conduct original, independent research and to publish their findings in peer-reviewed publications.

Fellowship Details:
Putnam Fellows are full-time employees of Harvard University during their tenure, with stipends of up to $53,000 per year depending on the duration of the fellowship, and are eligible for health insurance benefits. Modest support is available for research expenses and travel costs. The fellowship is typically awarded for 2 years, pending a satisfactory progress report at the end of the first year. Putnam Fellows are expected to be in full-time residence at the Arboretum and are provided office and research space.

The Putnam Fellowship is an independent post-doctoral position. As an independent scholar, Putnam Fellows have access to shared laboratories, resources, and interactions with fellow scientists, students and staff. It is not necessary to have a specific faculty host.

RESEARCH PROJECT: Genetic basis of hornwort carbon-concentrating
mechanisms.

Biophysical carbon concentrating mechanisms (CCMs) operating at the
single-cell level have evolved independently in some eukaryotic algae and
a single lineage of land plants, the hornworts. An essential component
for an efficient biophysical CCM is a pyrenoid, which represents a
specialized compartment inside chloroplasts that mainly comprise the
CO2-fixing enzyme RuBisCO. Hornworts with pyrenoids fix significantly
more carbon than their relatives without pyrenoids. Given the repeated
gains and losses of pyrenoids in hornworts during the last 50 million
years, we may assume that their assembly is potentially controlled by a
few master regulators of eco-evolutionary relevance. In a joint effort,
we will combine comparative -omics with reverse genetics tools to study
the genetics, function, and molecular basis of pyrenoid-based CCM in
hornwort plastids under different environmental conditions. Guided by
ultrastructure-based monitoring of the pyrenoid assembly in hornworts,
we aim to identify the genetic toolkit of biophysical CCM in hornworts
through two interdependent approaches: First, we aim to predict candidate
CCM components in silico though a set of homology searches that compare
the hornwort gene set with algal CCM component. Second, we employ an
exploratory gene and protein (co)expression profiling of isolated plastids
collected under low vs. high CO2 concentrations and under flooding. A
strength of our experimental design is that we contrast up to three
pairs of pyrenoid bearing and pyrenoid lacking hornwort species. Finally,
we will investigate pyrenoid functionality under various environmental
conditions. Specifically, we aim to conduct localization and functional
validation analyses for a core set of genes discovered in our CCM gene
prediction approaches. These experiments are possible through our recent
advances to establish Anthoceros agrestis and other hornwort species as a
tractable model system. Together, our collaborative project will not only
allow a comparison of the mechanisms of pyrenoid assembly between algae
and hornworts, but also reveal general principles and species-specific
innovations in the evolution of carbon-concentrating plastids. Above that,
focusing on and understanding the basis of land plant CCM instead of
only the algal form could eventually contribute to efficiently engineer
pyrenoid assembly and boost photosynthetic efficiency of crops. Relevant

This project is founded (initially for three years) by the German National
Science Foundation (DFG) MadLand (https://madland.science/) program to
Peter Szovenyi and Susann Wicke. The project requires collaborative work
including frequent visits to Germany and to the USA (Fay-Wei Li`s lab
at Cornell).

The Dept. of Systematic Botany hosts research groups working on
the evolutionary and ecological drivers of biodiversity, on the
macroevolution of plants, on plant-insect interactions/pollination,
on the evolution of mating systems, hybridization and speciation. The
Dept. of Plant and Microbial Biology hosts many groups working on plant
molecular and developmental biology, epigenetics, community genomics and
plant adaptation. Both institutes are housed in the beautiful Botanical
Gardens and host a diverse community of researchers in plant biology.

Ideal candidates will have an MSc in biology with a specialization in
evolution, developmental genetics and/or bioinformatics. This position
primarily involves reverse genetic, microscopy, gas exchange measurements
and bioinformatic work and requires advanced skills in handling, analyzing
and interpreting high-throughput next-generation sequencing data. Good
skills in assembling vectors, carrying out genetic transformations and
microscopy are also required. Previous experience with Chlamydomonas
pyrenoids is a plus but not necessary. In case not all these skills
are covered, the willingness to quickly acquire them is absolutely
necessary. The student will closely work together with the second
PhD student/postdoctoral associate in Germany on this grant. Students
should be willing to work both in the wet lab and in the office doing
computational work. The position is initially for three years. Selected
candidates will be enrolled in one of the two affiliated PhD schools in
evolution or plant sciences.

CLOSING DATE: The position is opened until filled, but all application

The Max-Planck-Institute for Plant Breeding Research (MPIPZ),
the University of Cologne and the Heinrich Heine University Düsseldorf

the International Max Planck Research School (IMPRS) on
“Understanding complex plant traits using computational and
evolutionary approaches” in Cologne and Düsseldorf, Germany.

The IMPRS is intended for highly motivated students with a strong
training in plant molecular sciences. The constellation of
participating institutions provides excellent conditions in the area
of plant sciences with expertise in molecular plant genetics, plant
biochemistry, cell biology, bioinformatics, computational genetics and
mathematical modelling of biological processes.

The training includes regular seminars, yearly retreats, supervision
by a thesis committee, and soft skill and practical courses on modern
laboratory techniques.

Master, Ph.D, Postdoctoral or Research Associate in Plant Molecular Genetics and Physiology (China)

Postgraduate research candidate positions (Master, Ph.D, Postdoctoral or Research Associate) in the field of Plant Molecular Genetics and Physiology are available in the Molecular Plant Physiology group at the Department of Biology, Zhejiang A & F University at Hangzhou and will be supervised by Prof. Bingsong Zheng. The group is using physiological, molecular and genomic tools to better understand how environment change will affect forest trees.

The successful candidate will work in the project screening tools for assessing adaptation of woody plant to environment and will examine woody plant as a model to investigate adaptation and acclimation of trees to environment change. Candidates are expected to work in an interdisciplinary research project, get involved in ongoing experiments, to assess genetic variation in trees from experiments and associate physiological and genetic data.

Candidate requirements

Candidates must hold or expect to obtain at least a bachelor, Master or Ph.D degree in the field of biology or a related discipline (plant physiology, plant breeding, molecular biology, forestry etc.) and should have strong writing skills. Candidates should (1) be proficient with one or more languages (English, simple oral, listen and written Chinese if possible); (2) have an interest in plant molecular genetics and physiology.

Linking the genome to the niche in the mega-diverse genus Begonia
PhD scholarship

Abstract
Begonias are one of the largest groups of flowering plants in the world,
with nearly 2000 species. They are very successful at colonising deeply
shaded habitats in the tropical forest understory, often growing where
few other plants can survive.  Why are there 2000 species, and how are
they different? Answering this question will give us insights into why
the tropics hold so much of the world's biodiversity. To what extent
do begonias differ in tolerating deep shade, which is a very important
part of their strategy for survival? To answer this we need to compare
the light levels that a number of different species get from dawn to
dusk by measuring it in the forest where they grow. There may be other
important factors in the habitat that may differ from site to site,
such as temperature, humidity and soil characteristics, so we also need
to measure those. From this data we can see how much the conditions
each species grows in differ. What would make each species do better
under different conditions? To answer this we need to look at their
ecophysiology, for example examining photosynthetic efficiency, how the
leaf surface might vary in order to capture light of different intensity
or angle, or how the stomata are arranged and controlled. To complete
the picture of how begonia species differ, we can use our database of
begonia genes to identify genes variable between species, focusing on
those which may be involved in ecophysiological traits. We can sequence
these from the same species we have habitat and ecophysiology data
for to determine if  the sequence variation is linked to adaptation to
particular niches. There is the potential to use interspecific genetics
or transgenics to  further investigate functional links between particular
genes and niche adaptation.

Applicants should be highly motivated with an excellent academic record,
with a first degree in a relevant subject of at least upper second
or equivalent. The project will require competence in molecular lab
skills and the ability to withstand several months over the course of
the project in the field in Indonesia.

The studentship is funded by the M. L. MacIntyre Begonia Trust, and is
open to applicants liable for tuition fees at the Home and EU rate. The
stipend is £13590 per annum. The start date will be in September-October
2013, with a closing date for applications of August 30th 2013. Applicants

applicant will be based at the Royal Botanic Garden Edinburgh, with the

Postdoc position in Plant Evolutionary Genetics

Postdoc position in Plant Evolutionary Genetics at Newcastle University,
UK.  We are looking for an enthusiastic postdoc for a 3-year NERC-funded
research project entitled: 'Inter-genomic conflict in gynodioecy and
its effects on molecular evolution of mitochondrial genomes in the
genus Plantago'.

The applicant will be based in Kirsten Wolff's group in the School
of Biology, Newcastle University, and will collaborate closely with
Deborah Charlesworth's group at the Institute of Evolutionary Biology,
University of Edinburgh, where an additional postdoc will be part of
the same project.

This project will use molecular evolutionary approaches to investigate
the maintenance of cytonuclear polymorphisms in gynodioecious plants
in the genus Plantago, to test whether an 'arms race' situation
prevails, versus long-term balancing selection. In the project we
will be obtaining and analysing DNA sequences of multiple nuclear
and mitochondrial genes, to compare diversity, and test for long-term
balancing selection. Non-gynodioecious species will be studied, as well as
gynodioecious ones, to provide outgroups for sequence analyses and to test
whether the high diversity expected under long-term balancing selection
is confined to the gynodioecious species. The project also includes
testing for paternal inheritance/leakage by testing for heteroplasmy in
natural populations of P. lanceolata, using analyses of mitochondrial
DNA sequence haplotypes and studies of offspring of reciprocal crosses
to assess the possibility that within-species polymorphism could explain
the highly elevated mutation rates estimated in Plantago lineages.

The work will be divided between Edinburgh and Newcastle. For example,
the Edinburgh group will concentrate on the nuclear genes, while the
Newcastle group will obtain the mitochondrial genes, while other work
is shared. Field work and greenhouse work will predominantly be done by
the Newcastle postdoc.

Applicants must have completed their PhD before starting the position.
Applicants should have some experience with molecular genetic techniques,
and an interest in population genetics.
